Drug intelligence / Profile preview
PA-T-CAT mAb is an engineered recombinant fusion protein and monoclonal antibody candidate developed by Omeros Corporation in collaboration with the University of Cambridge. It is designed using Omeros' proprietary Targeted Complement Activation Therapy (T-CAT) platform to treat severe, multidrug-resistant (MDR) *Pseudomonas aeruginosa* infections, such as pneumonia and sepsis. The antibody component of PA-T-CAT mAb specifically targets the *P. aeruginosa* flagellin type B (Fla-B) antigen. The heavy chains of the antibody are fused to a truncated, enzymatically active form of human complement component 1s (C1s), containing the CCPI, CCPII, and serine protease (SP) domains. This design allows PA-T-CAT mAb to bypass the classical complement pathway's requirement for the C1 initiation complex, directly cleaving complement components C4 and C2 on the bacterial surface. This localized activation drives robust C3b deposition, membrane attack complex (MAC) assembly, and complement-mediated bacterial lysis and clearance, overcoming bacterial immune evasion mechanisms.
