PA3-17 is an autologous chimeric antigen receptor (CAR) T-cell therapy specifically engineered to target CD7, a cell surface glycoprotein highly expressed on malignant T cells in diseases such as relapsed/refractory T-lymphoblastic leukemia/lymphoma (r/r T-ALL/LBL) and other CD7-positive hematologic malignancies. Developed by PersonGen BioTherapeutics, PA3-17 uses a novel approach to prevent fratricide among CAR-T cells by blocking the expression of CD7 on the surface of the therapeutic T cells through an anti-CD7 protein expression blocker (PEBL). This allows for effective expansion and persistence of CAR-T cells targeting malignant CD7-expressing lymphocytes. Clinical studies have demonstrated encouraging safety and efficacy profiles in patients with r/r T-ALL/LBL, with high overall response rates and manageable toxicity[1][2][5][6].
