PA32540 + celecoxib is a combination of two pharmaceutical products. - **PA32540** is a coordinated-delivery tablet that combines enteric-coated aspirin (acetylsalicylic acid) 325 mg with immediate-release omeprazole 40 mg. It is designed to provide secondary cardiovascular protection while reducing the risk of upper gastrointestinal complications such as gastric ulcers associated with long-term aspirin use[7][4]. Aspirin acts as an antiplatelet agent by irreversibly inhibiting cyclooxygenase-1 (COX-1), thereby reducing thromboxane A2 production and platelet aggregation. Omeprazole is a proton pump inhibitor that suppresses gastric acid secretion by inhibiting the H+/K+ ATPase in gastric parietal cells. - **Celecoxib** is a selective nonsteroidal anti-inflammatory drug (NSAID) classified as a COX-2 inhibitor. It reduces pain and inflammation by selectively inhibiting cyclooxygenase-2, which decreases prostaglandin synthesis at sites of inflammation[5][6][8]. Celecoxib is used for conditions such as osteoarthritis, rheumatoid arthritis, ankylosing spondylitis, juvenile rheumatoid arthritis in children over 2 years old, acute pain including menstrual pain and post-surgical pain, and to reduce polyps in familial adenomatous polyposis[1][2]. This combination may be considered for patients requiring both cardiovascular protection from aspirin and anti-inflammatory or analgesic effects from celecoxib but who are at increased risk for gastrointestinal side effects.
