Overview

PA5691 is a preclinical-stage sustained-release ocular implant developed by PolyActiva for the treatment of diabetic macular edema (DME). The implant utilizes PolyActiva's proprietary polymer-drug conjugate technology, where the active pharmaceutical ingredient, dexamethasone, is chemically linked to a biodegradable polymer backbone. This design allows for a zero-order release profile, providing a constant dose of the corticosteroid directly to the retina over an extended period. By delivering dexamethasone in a controlled manner, PA5691 aims to reduce inflammation and vascular leakage associated with DME while potentially minimizing the side effects and injection frequency typically associated with standard intravitreal steroid treatments. The implant is designed to biodegrade completely once the drug payload has been fully released.
