Overview

PA65-2 is a monoclonal antibody (mAb) that specifically targets the human clathrin heavy chain (CHC). Identified by researchers at Academia Sinica, PA65-2 binds to tumor cells and tumor-associated blood vessels while showing no significant binding to normal tissues. The mechanism of action involves the inhibition of CHC, which normally functions to stabilize the transcription factor subunit HIF-1α and facilitate its nuclear translocation. By disrupting this interaction, PA65-2 reduces the expression of vascular endothelial growth factor (VEGF), thereby inhibiting tumor angiogenesis and growth. Preclinical studies in pancreatic adenocarcinoma xenograft models have demonstrated its potential as a therapeutic agent and prognostic indicator.
