Overview

PAC-336 is a selective small-molecule muscarinic acetylcholine receptor M4 (M4) agonist under development by Pace Pharmaceuticals for the treatment of schizophrenia. The compound is designed to selectively activate the M4 receptor, a G protein-coupled receptor primarily coupled to Gi/o proteins that is highly expressed in the striatum and cortex, where it regulates dopaminergic and cholinergic neurotransmission. By selectively targeting the M4 receptor, PAC-336 aims to address the positive, negative, and cognitive symptoms of schizophrenia while avoiding the side effects associated with non-selective muscarinic agonists. PAC-336 has completed the discovery phase and is undergoing IND-enabling studies, with plans to enter clinical testing.
