Overview

Pacibekitug is a long-acting, fully human monoclonal antibody that targets interleukin 6 (IL-6), a key proinflammatory cytokine involved in the pathogenesis of many autoimmune and inflammatory diseases. It exhibits best-in-class potential with properties such as a naturally long half-life, low immunogenicity, and high binding affinity to IL-6. Pacibekitug is administered subcutaneously and is being developed for infrequent dosing regimens (as little as once every quarter). The drug was originally developed by Pfizer (as PF‑04236921) and is now being advanced by Tourmaline Bio for indications including thyroid eye disease (TED/Graves' ophthalmopathy), atherosclerotic cardiovascular disease (ASCVD), chronic kidney disease, and other inflammation-driven conditions. Clinical trials have demonstrated deep inhibition of the IL‑6 signaling pathway with relatively low drug exposure[1][2][3][4][5][8].
