Overview

Paclimer is a controlled-release, biodegradable formulation of paclitaxel encapsulated within polilactofate microspheres. Designed for local and regional administration—such as intraperitoneal, intratumoral, or intracranial injection—the system allows for the sustained release of paclitaxel over several weeks to months. This delivery method aims to maintain high therapeutic concentrations of the drug at the tumor site while keeping systemic plasma levels low to reduce toxicity. The active ingredient, paclitaxel, is a taxane that stabilizes microtubules, thereby inhibiting mitosis and leading to cell death. Development of Paclimer, which reached Phase I clinical trials for ovarian and peritoneal cancers, was ultimately suspended by the manufacturer.
