**Paclitaxel** is a small-molecule taxane microtubule stabilizer. It binds beta-tubulin in polymerized microtubules, promotes microtubule assembly, and prevents depolymerization, disrupting mitotic-spindle dynamics and cell division. In the University of Rome Tor Vergata-sponsored PACUS trial, paclitaxel was evaluated as a locally delivered antiproliferative agent after femoropopliteal angioplasty for critical limb ischemia; catheter-delivered high-intensity, low-frequency ultrasound and temporary flow occlusion were used to enhance arterial-wall uptake. This was a delivery-strategy study of paclitaxel rather than a distinct new molecular entity.
