Overview

The combination of **paclitaxel**, **carboplatin**, and **vorinostat** is an investigational chemotherapeutic regimen used in clinical trials for cancer. Paclitaxel is a microtubule-stabilizing agent (taxane class), carboplatin is a DNA-damaging platinum agent, and vorinostat is a histone deacetylase (HDAC) inhibitor. Vorinostat enhances the anticancer effects of platinum compounds and taxanes by inducing chromatin remodeling, increasing DNA damage, and modulating gene expression, both through histone and nonhistone-mediated mechanisms. Clinically, this combination has been evaluated in phase II studies in patients with **advanced non-small-cell lung cancer (NSCLC)** and **platinum-sensitive recurrent ovarian, peritoneal, or fallopian tube carcinoma**, demonstrating improved response rates, tolerable safety profiles, and promising survival outcomes compared to carboplatin plus paclitaxel alone[1][2][3][4].
