Overview

Paclitaxel + anthracycline refers to a chemotherapy regimen combining the taxane class drug paclitaxel with an anthracycline (most commonly doxorubicin or epirubicin). This combination is primarily used in the treatment of metastatic and advanced breast cancer. Paclitaxel stabilizes microtubules and inhibits cell division, while anthracyclines intercalate DNA and inhibit topoisomerase II, leading to DNA damage and apoptosis. The regimen has demonstrated high response rates in clinical trials but is associated with risks of cardiotoxicity—especially when doxorubicin is used—necessitating careful cardiac monitoring. Substituting epirubicin for doxorubicin can reduce cardiac toxicity while maintaining efficacy[1][3][4].
