Overview

Paclitaxel + carboplatin is a combination chemotherapy regimen that was investigated by the All India Institute of Medical Sciences (AIIMS) for the treatment of cervical cancer. Paclitaxel is a taxane-based small molecule that works by binding to the beta-subunit of tubulin, thereby stabilizing microtubules and preventing their depolymerization. This action arrests the cell cycle in the G2/M phase and induces apoptosis. Carboplatin is a platinum-based antineoplastic agent that forms reactive platinum complexes, which create intra-strand and inter-strand DNA cross-links. These cross-links interfere with DNA replication and transcription, leading to cell death. The AIIMS clinical trial (NCT01267838) specifically evaluated this combination in patients with metastatic, recurrent, or persistent cervical cancer, comparing the efficacy and safety of weekly versus three-weekly dosing schedules. It has also been studied as a dose-dense neoadjuvant chemotherapy approach for locally advanced cervical cancer.
