The combination of paclitaxel, carboplatin, and megestrol acetate is an investigational chemo-hormonal regimen evaluated by the University of Alabama at Birmingham for the treatment of advanced-stage or recurrent endometrial cancer. This regimen integrates two standard cytotoxic chemotherapy agents with a hormonal therapy component. Paclitaxel is a taxane that stabilizes microtubules, thereby inhibiting cell division, while carboplatin is a platinum-based agent that induces DNA cross-linking and subsequent apoptosis. Megestrol acetate is a synthetic progestin that exerts anti-estrogenic effects on the endometrium, potentially sensitizing tumor cells or providing additive anti-tumor activity. This combination was primarily studied in a Phase II clinical trial (NCT00114608) to determine if adding hormonal modulation to standard chemotherapy could improve outcomes in patients with FIGO Stage III-IV or recurrent disease.
