Overview

This therapeutic regimen refers to a specific treatment protocol developed and evaluated by the Radiation Therapy Oncology Group (RTOG), most notably in the RTOG 0229 clinical trial. The regimen is designed for patients with locally advanced (Stage III) non-small cell lung cancer (NSCLC) and consists of neoadjuvant concurrent chemoradiotherapy followed by surgical resection and consolidation chemotherapy. The chemotherapy component utilizes paclitaxel, a microtubule-stabilizing agent that induces cell cycle arrest by preventing mitotic spindle disassembly, and carboplatin, a platinum-based antineoplastic agent that forms DNA cross-links to inhibit replication. These agents are administered alongside high-dose radiation therapy (typically 61.2 Gy) to maximize local tumor shrinkage and systemic control before surgery. Following resection, patients receive consolidation therapy with the same chemotherapy doublet to eliminate residual micrometastatic disease.
