Overview

This combination induction therapy regimen, evaluated in a Phase 2 clinical trial led by Dr. Susanne Arnold at the University of Kentucky, is designed for the treatment of locally advanced squamous cell carcinoma of the head and neck (SCCHN). The protocol involves the administration of the taxane chemotherapy agent paclitaxel and the platinum-based agent carboplatin in conjunction with low-dose fractionated radiation therapy (LDFRT). The LDFRT component (typically administered in 50-80 cGy fractions) is utilized as a 'chemoenhancer' to exploit the biological phenomenon of hyper-radiation sensitivity, which increases the cytotoxic efficacy of the systemic chemotherapy agents. This approach aims to achieve high response rates at both the primary tumor site and involved lymph nodes prior to definitive surgical or full-dose radiation treatment.
