Overview

Paclitaxel + cisplatin is a combination chemotherapy regimen widely used in the treatment of various solid tumors, including non-small cell lung cancer (NSCLC), ovarian cancer, cervical cancer, and esophageal cancer. Paclitaxel is a microtubule-stabilizing agent that binds to tubulin and promotes microtubule assembly while inhibiting disassembly, causing cell cycle arrest at the G2/M phase and inducing apoptosis. Cisplatin is a platinum-based chemotherapeutic that forms DNA crosslinks (primarily intrastrand) leading to DNA damage and subsequent apoptosis. The combination exploits synergistic cytotoxicity by targeting different mechanisms within tumor cells. This regimen is typically administered intravenously in cycles every 2–3 weeks depending on indication and patient tolerance[1][2][3][5]. It is considered standard first-line therapy for advanced NSCLC (especially squamous subtype), recurrent or metastatic cervical cancer, advanced ovarian carcinoma, and as part of concurrent chemoradiotherapy protocols for esophageal carcinoma[1][2][3][4].
