Overview

This induction chemotherapy regimen, investigated by Barretos Cancer Hospital (also known as Hospital de Amor), is a combination therapy consisting of paclitaxel and cisplatin for the treatment of locally advanced head and neck squamous cell carcinoma (HNSCC). Paclitaxel is a taxane that stabilizes microtubules, preventing their depolymerization and leading to cell cycle arrest, while cisplatin is a platinum-based agent that forms covalent DNA cross-links, interfering with replication and transcription. The protocol typically involves three cycles of induction with these agents prior to definitive chemoradiotherapy. A Phase 3 trial (NCT01133002) was conducted to evaluate if this induction strategy improves overall survival compared to standard chemoradiotherapy alone in patients with stage III-IV HNSCC.
