Paclitaxel + cisplatin + PD-1 inhibitor (TP + PD-1) is a chemo-immunotherapy combination regimen used for the first-line treatment of advanced or metastatic esophageal squamous cell carcinoma (ESCC). The regimen combines two cytotoxic chemotherapy agents—paclitaxel, a taxane that stabilizes microtubules to inhibit mitosis, and cisplatin, a platinum-based agent that causes DNA cross-linking—with a monoclonal antibody targeting the Programmed Death-1 (PD-1) receptor. By blocking the PD-1/PD-L1 pathway, the PD-1 inhibitor restores T-cell mediated anti-tumor activity. This combination is being studied in various clinical settings, including biomarker-guided umbrella trials led by institutions like Sun Yat-sen University, to improve survival outcomes compared to standard chemotherapy alone.
