This is a combination regimen consisting of four agents: paclitaxel (a microtubule-stabilizing chemotherapeutic), cisplatin (a platinum-based DNA crosslinking agent), TQB2450 (a humanized anti-PD-L1 monoclonal antibody immune checkpoint inhibitor), and anlotinib (an oral small molecule multitargeted tyrosine kinase inhibitor primarily targeting vascular endothelial growth factor receptors 1–3, among others). The combination is under investigation as a first-line treatment for advanced cancers such as esophageal squamous cell carcinoma and biliary tract cancer. Paclitaxel disrupts mitosis by stabilizing microtubules; cisplatin induces DNA damage leading to apoptosis; TQB2450 blocks PD-L1 to enhance antitumor immune response; and anlotinib inhibits angiogenesis and tumor proliferation via VEGFR inhibition. This multi-agent approach aims to combine cytotoxic, immunotherapeutic, and antiangiogenic mechanisms for synergistic efficacy in advanced solid tumors[1][2][4].
