paclitaxel + ramucirumab

Paclitaxel + ramucirumab is a combination therapy used primarily as a second-line treatment for patients with advanced or metastatic gastric or gastroesophageal junction (GEJ) adenocarcinoma whose disease has progressed after prior fluoropyrimidine- or platinum-containing chemotherapy. Paclitaxel is a microtubule inhibitor that disrupts cell division, while ramucirumab is a fully human IgG1 monoclonal antibody targeting vascular endothelial growth factor receptor 2 (VEGFR-2), thereby inhibiting angiogenesis and tumor blood supply. The combination has demonstrated improved overall survival and progression-free survival compared to paclitaxel alone in this setting, as shown in the pivotal phase III RAINBOW trial[1][2][7][10]. This regimen is FDA-approved for this indication and recommended by clinical guidelines.
