A combination chemotherapy regimen consisting of paclitaxel (a taxane), tegafur/uracil (UFT, an oral fluoropyrimidine), and leucovorin (LV, folinic acid). Paclitaxel acts as an antimitotic agent by stabilizing microtubules. Tegafur is a prodrug that converts to 5-fluorouracil (5-FU) in the body, while uracil competitively inhibits dihydropyrimidine dehydrogenase (DPD) to prevent rapid breakdown of 5-FU. Leucovorin enhances the cytotoxic effects of 5-FU by stabilizing the binding of 5-FU metabolites to thymidylate synthase, thereby inhibiting DNA synthesis in cancer cells.
