Overview

This combination therapy regimen, evaluated in the HISTORIA trial, is a conversion treatment for patients with gastric or esophagogastric junction cancer and peritoneal metastasis. The protocol involves Hyperthermic Intraperitoneal Chemotherapy (HIPEC) using paclitaxel to target peritoneal disease, followed by systemic therapy. The systemic component includes the PD-1 inhibitor tislelizumab, the SOX chemotherapy regimen (oxaliplatin and the oral fluoropyrimidine S-1), and the HER2-targeted monoclonal antibody trastuzumab for patients with HER2-positive tumors. This multimodal approach combines cytotoxic chemotherapy, immunotherapy, and targeted therapy to maximize tumor response and facilitate radical surgical resection.
