paclitaxel + topotecan + estramustine is a combination chemotherapy regimen investigated by the Fox Chase Cancer Center for the treatment of metastatic hormone-refractory prostate cancer (mHRPC). The regimen utilizes three agents with distinct but complementary mechanisms: paclitaxel, a taxane that stabilizes microtubules to induce mitotic arrest; topotecan, a camptothecin analog that inhibits DNA topoisomerase I to cause lethal DNA damage; and estramustine phosphate, a conjugate of estradiol and nitrogen mustard that disrupts microtubule dynamics by binding to microtubule-associated proteins. This multi-targeted approach was evaluated in Phase II clinical trials to improve outcomes in patients whose prostate cancer had become resistant to standard hormonal therapies.
