Overview

Paclitaxel + toremifene is an investigational combination therapy consisting of two distinct drugs used primarily in the treatment of metastatic breast cancer. Paclitaxel is a taxane-class chemotherapeutic agent that stabilizes microtubules, thereby inhibiting cell division and inducing apoptosis in rapidly dividing tumor cells. Toremifene is a selective estrogen receptor modulator (SERM) that acts as an antiestrogen by competitively binding to estrogen receptors, blocking the proliferative action of estrogen on breast cancer cells. The rationale for combining these agents includes both direct cytotoxic effects from paclitaxel and endocrine modulation from toremifene, with additional evidence suggesting that toremifene may help overcome P-glycoprotein-mediated drug resistance associated with paclitaxel therapy[1][2][3]. Clinical studies indicate this combination has been evaluated for safety and efficacy in patients with previously treated metastatic breast cancer, showing tolerability and similar pharmacokinetic profiles compared to monotherapy[2][3].
