Overview

A two-drug anticancer combination of albumin-bound paclitaxel (nab-paclitaxel) and sorafenib. Nab-paclitaxel is a solvent-free, nanoparticle albumin-bound formulation of paclitaxel that stabilizes microtubules to inhibit mitosis, while sorafenib is an oral multikinase inhibitor targeting RAF kinases and receptor tyrosine kinases involved in angiogenesis and tumor proliferation, including VEGFR and PDGFR. Preclinical studies show synergistic anti-tumor effects of paclitaxel with sorafenib, including enhanced suppression of cancer stem cell properties and increased apoptosis; early clinical development has evaluated Abraxane with continuous sorafenib dosing in solid tumors such as breast cancer as first-line therapy in phase II settings. Nab-paclitaxel is approved as a single agent for several cancers, but this specific combination does not have regulatory approval and has been explored in trials rather than established practice.
