Overview

Paclitaxel gallium transferrin (PGT) is an investigational CD71-targeted protein complex that combines the microtubule-stabilizing taxane paclitaxel with the metal-based anticancer agent gallium, both associated with a human transferrin scaffold, to selectively deliver therapy to tumors overexpressing transferrin receptor 1 (CD71/TFR1). Paclitaxel inhibits mitosis by binding β-tubulin and stabilizing microtubules, leading to G2/M arrest and apoptosis, while gallium, a trivalent metal with iron-mimetic and antiproliferative properties, provides a second non–cross-resistant cytotoxic mechanism.[1][5][8][4] Transferrin serves as a natural ligand for CD71, enabling targeted uptake into CD71-high cancer cells and potentially improving therapeutic index versus solvent-based and albumin-bound paclitaxel by enhancing tumor delivery and adding gallium-mediated antitumor activity.[4][3]
