Paclitaxel liposome + cisplatin is a combination chemotherapy regimen used primarily for the treatment of advanced or metastatic non-small cell lung cancer (NSCLC) and lung squamous cell carcinoma. Paclitaxel, formulated as a liposome (notably under the brand name Lipusu), is designed to improve drug delivery and reduce toxicity compared to conventional formulations. Cisplatin, sometimes also delivered in a liposomal form (Lipoplatin), is a platinum-based chemotherapeutic agent that forms DNA crosslinks, leading to apoptosis in rapidly dividing cells. The combination leverages the microtubule-stabilizing action of paclitaxel with the DNA-damaging effects of cisplatin for synergistic antitumor activity. Clinical studies have shown that this combination can be less toxic than traditional regimens using non-liposomal drugs while maintaining efficacy[1][2][5]. Paclitaxel acts by stabilizing microtubules and inhibiting their depolymerization, thereby blocking mitosis[3]. Cisplatin binds to DNA and causes crosslinking which inhibits DNA synthesis and function, ultimately triggering apoptosis[4][8].
