Oncofid-P-M (paclitaxel obaluronate) is an innovative polymer-drug conjugate consisting of the chemotherapeutic agent paclitaxel covalently linked to hyaluronic acid. Developed by Fidia Farmaceutici, it is designed for the loco-regional treatment of malignant pleural mesothelioma. The hyaluronic acid component of the conjugate specifically targets and binds to CD44, a cell-surface receptor overexpressed on mesothelioma cells, thereby facilitating receptor-mediated endocytosis and increasing the intracellular concentration of paclitaxel while minimizing systemic exposure and toxicity. Once inside the cell, paclitaxel exerts its cytotoxic effect by binding to and stabilizing microtubules, inhibiting cell division and inducing apoptosis. Oncofid-P-M is administered intrapleurally and is currently being evaluated in Phase I clinical trials for recurrent or progressive pleural mesothelioma.
