Drug intelligence / Profile preview
Paclitaxel polymer micelles is a nanoparticle-based formulation of the taxane chemotherapy agent paclitaxel, designed to enhance the drug's therapeutic index. By encapsulating paclitaxel within amphiphilic block copolymer micelles (typically PEG-PDLLA), the formulation eliminates the need for Cremophor EL and ethanol, which are responsible for the severe hypersensitivity reactions associated with traditional paclitaxel formulations. This micellar delivery system improves the solubility of paclitaxel, allows for higher dose intensity and shorter infusion times without the need for extensive corticosteroid premedication, and leverages the enhanced permeability and retention (EPR) effect to increase drug concentration within tumor tissues. In clinical research led by Shanghai 6th People's Hospital (notably the MAGNATE-S trial), this formulation is being evaluated in combination with gemcitabine and targeted tyrosine kinase inhibitors (such as Lenvatinib or Anlotinib) for the treatment of advanced bone and soft tissue sarcomas that have progressed after standard first-line therapy.
