Overview

This combination therapy regimen consists of a novel nanoparticle formulation of paclitaxel (polymer micelles), the antimetabolite gemcitabine, and the multi-kinase inhibitor lenvatinib. It is being investigated by Shanghai 6th People's Hospital in the Phase 2 MAGNATE-S trial for the treatment of advanced, unresectable, or metastatic bone sarcomas that have progressed after first-line therapy. Paclitaxel polymer micelles are designed to improve solubility and reduce hypersensitivity reactions compared to traditional paclitaxel. Gemcitabine inhibits DNA synthesis by acting as a nucleoside analog, while lenvatinib targets multiple tyrosine kinases including VEGFR1-3, FGFR1-4, PDGFRα, KIT, and RET to inhibit angiogenesis and tumor cell proliferation. In the MAGNATE-S study, this specific combination is primarily allocated to patients with bone sarcoma, while a similar regimen substituting lenvatinib with anlotinib is used for soft tissue sarcoma.
