Overview

paclitaxel polymer micelles + trastuzumab + pertuzumab + adebelimab is an investigational combination therapy being evaluated as a first-line treatment for patients with advanced HER2-positive breast cancer. This regimen integrates chemotherapy, dual HER2-targeted therapy, and immune checkpoint inhibition. Paclitaxel polymer micelles are a nanoparticle formulation of the microtubule-stabilizing agent paclitaxel, designed to enhance solubility and allow for higher dosing with improved safety compared to traditional solvent-based formulations. Trastuzumab and pertuzumab are monoclonal antibodies that target distinct domains of the human epidermal growth factor receptor 2 (HER2), providing a synergistic blockade of HER2 signaling and promoting antibody-dependent cellular cytotoxicity (ADCC). Adebelimab (SHR-1316) is a humanized monoclonal antibody targeting programmed death-ligand 1 (PD-L1), which acts to restore the anti-tumor activity of T cells. The combination aims to leverage the cytotoxic effects of chemotherapy and the targeted action of anti-HER2 antibodies alongside the immune-potentiating effects of PD-L1 inhibition.
