Paclitaxel polymer micelles for injection + cisplatin + sintilimab is an investigational combination regimen being evaluated as a neoadjuvant (pre-surgical) therapy for patients with resectable, locally advanced esophageal squamous cell carcinoma (ESCC). The regimen combines three active components: (1) paclitaxel polymeric micelles for injection (Pm-Pac), a nanoparticle formulation of the taxane chemotherapy agent paclitaxel that uses polymeric micelles to enhance tumor permeability and retention while avoiding the need for steroid premedication; (2) cisplatin, a platinum-based chemotherapy agent that causes DNA cross-linking and induces cancer cell apoptosis; and (3) sintilimab, a fully human IgG4 monoclonal antibody targeting the PD-1 (programmed cell death protein 1) receptor to block the PD-1/PD-L1 pathway and restore anti-tumor T-cell activity. This combination is designed to leverage the immunogenic and cytotoxic effects of nanoformulated chemotherapy alongside PD-1 checkpoint blockade to achieve pathological responses prior to surgical resection.
