Overview

A combination drug product consisting of paclitaxel formulated as polymeric micelles and gemcitabine, co-administered for synergistic antineoplastic activity primarily in pancreatic cancer. The paclitaxel is encapsulated within nanosized polymeric micelles, enhancing its solubility, stability, and tumor accumulation compared to conventional formulations. Gemcitabine, a nucleoside analog, acts as a DNA synthesis inhibitor. Co-formulation or co-administration leverages the controlled and preferential delivery properties of micelles for paclitaxel, and the established cytotoxicity of gemcitabine, resulting in improved therapeutic efficacy and reduced toxicity. This combination is primarily being tested as a first-line therapy in metastatic pancreatic cancer, with ongoing phase 3 clinical evaluation.
