This combination therapy consists of paclitaxel polymersomes, carboplatin, and adebelimab, currently being evaluated as a neoadjuvant treatment for resectable mucosal melanoma. Paclitaxel polymersomes utilize a nanoparticle delivery system (polymersomes) to provide controlled release of paclitaxel, a taxane that stabilizes microtubules and inhibits mitosis. Carboplatin is a platinum-based chemotherapeutic agent that induces DNA interstrand and intrastrand cross-links, leading to apoptosis. Adebelimab (SHR-1316) is a humanized IgG4 monoclonal antibody targeting Programmed Death-Ligand 1 (PD-L1), which prevents PD-L1 from binding to PD-1 and B7.1 receptors on T cells, thereby enhancing the immune system's ability to identify and destroy tumor cells. The regimen is being studied by Sun Yat-sen University in a Phase 2 clinical trial to assess pathological response rates in patients with resectable mucosal melanoma.
