Nanonext is an approved generic formulation of nanoparticle albumin-bound paclitaxel (nab-paclitaxel), marketed by Perin Healthcare. It is an injectable suspension containing paclitaxel protein-bound particles, where the active chemotherapeutic agent paclitaxel is non-covalently bound to human albumin nanoparticles. This solvent-free formulation avoids the use of Cremophor EL, a vehicle used in conventional paclitaxel formulations that is associated with severe hypersensitivity reactions and requires corticosteroid premedication. Paclitaxel is a microtubule inhibitor that binds to tubulin, promoting microtubule assembly and stabilizing them against depolymerization, which disrupts mitotic spindle dynamics and leads to cell cycle arrest and apoptosis. Nanonext is indicated for the treatment of metastatic breast cancer, advanced non-small cell lung cancer (NSCLC), and metastatic adenocarcinoma of the pancreas.
