Overview

Pacmilimab is a recombinant, human monoclonal antibody developed as a Probody therapeutic targeting programmed cell death ligand 1 (PD-L1). It is engineered to be conditionally activated in the tumor microenvironment by tumor-associated proteases, remaining predominantly masked and inert in healthy tissues. This design aims to restrict immune checkpoint inhibition activity to tumors, thereby reducing off-tumor toxicity compared with conventional anti-PD-L1 antibodies. Pacmilimab acts by blocking PD-L1, leading to enhanced T lymphocyte activation and antitumor immune responses. It has demonstrated antitumor activity and a favorable safety profile in early-phase clinical trials for various advanced solid tumors[2][3][4][5].
