Overview

Pacmilimab + praluzatamab ravtansine is a combination of two conditionally activated antibody-based cancer therapeutics developed by CytomX Therapeutics. Pacmilimab (CX-072) is a Probody therapeutic targeting PD-L1, designed to be activated specifically in the tumor microenvironment by tumor-associated proteases, thereby reducing systemic toxicity. Praluzatamab ravtansine (CX-2009) is a conditionally activated antibody-drug conjugate (ADC) that targets CD166 and delivers the cytotoxic agent DM4 via a protease-cleavable linker and peptide mask. This combination was evaluated in clinical trials for advanced triple-negative breast cancer (TNBC), aiming to leverage immune checkpoint inhibition with targeted cytotoxic delivery[1][3][5]. The development of this combination in TNBC was discontinued after failing to meet efficacy endpoints in Phase 2 studies[5][6].
