Drug intelligence / Profile preview
PAL-119 is an orally administered, selective small molecule inhibitor of dual-specificity tyrosine-phosphorylation-regulated kinase 1A (DYRK1A) under development by Paulex Bio. DYRK1A acts as a molecular brake on human pancreatic beta cell proliferation by phosphorylating and removing the transcription factor NFAT (nuclear factor of activated T-cells) from the nucleus. By inhibiting DYRK1A, PAL-119 restores NFAT nuclear localization and transcriptional activity, thereby promoting beta cell regeneration, differentiation, and improved insulin production. PAL-119 is being evaluated in Phase 1/2a clinical trials for the treatment of type 1 and type 2 diabetes, aiming to restore endogenous insulin production and potentially offer insulin independence.
