Overview

Palacaparib (AZD9574) is an investigational, highly brain-penetrant, next‑generation small‑molecule inhibitor of poly(ADP‑ribose) polymerase 1 (PARP1) being developed by AstraZeneca for the treatment of solid tumors, including those with defects in homologous recombination DNA repair. Preclinical characterization shows potent and selective inhibition of PARP1 with robust PARP‑DNA trapping and the ability to cross the blood–brain barrier, supporting exploration in tumors of the central nervous system and other indications where CNS penetration is advantageous. Palacaparib is currently in early‑phase clinical development, including a phase 2 trial across multiple solid tumor types such as endometrial, ovarian, prostate, glioma, pancreatic, and non‑small cell lung cancers.
