palbociclib + pioglitazone

Palbociclib is a small molecule inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6), developed by Pfizer, and is primarily used in combination with other agents for the treatment of hormone receptor-positive, HER2-negative advanced or metastatic breast cancer. It works by blocking CDK4/6 activity, thereby preventing phosphorylation of the retinoblastoma protein (Rb) and halting cell cycle progression from G1 to S phase, which inhibits cancer cell proliferation[2][5][6]. Pioglitazone is an oral antidiabetic agent from the thiazolidinedione class that acts as an agonist at peroxisome proliferator–activated receptor gamma (PPARγ), improving insulin sensitivity in patients with type 2 diabetes mellitus[3][4]. The combination "palbociclib + pioglitazone" refers to co-administration of these two drugs; there is no evidence this represents a fixed-dose combination product or a unique pharmaceutical entity.
