Overview

Palbociclib + talazoparib is a combination therapy investigated for the treatment of advanced solid tumors, particularly HR+/HER2- metastatic breast cancer. Palbociclib is a selective inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6), which regulates the cell cycle by preventing the transition from the G1 to the S phase. Talazoparib is a potent inhibitor of poly(ADP-ribose) polymerase (PARP) enzymes, specifically PARP1 and PARP2, which are essential for DNA repair. By inhibiting PARP and trapping the enzyme on DNA, talazoparib induces double-strand breaks that lead to apoptosis in cells with compromised DNA repair mechanisms, such as those with homologous recombination deficiency (HRD). The combination of a CDK4/6 inhibitor and a PARP inhibitor is hypothesized to provide synergistic anti-tumor activity by simultaneously disrupting cell cycle progression and DNA damage response pathways.
