Palbociclib + taselisib is an investigational combination therapy consisting of two small molecule inhibitors: palbociclib, a selective cyclin-dependent kinase 4/6 (CDK4/6) inhibitor, and taselisib, a selective phosphatidylinositol 3-kinase (PI3K) inhibitor. This combination has been studied primarily in advanced solid tumors and breast cancer with PIK3CA mutations. Palbociclib inhibits CDK4/6 to block cell cycle progression from G1 to S phase, while taselisib targets the PI3K pathway involved in cell growth and survival. The combination aims to overcome resistance mechanisms seen with single-agent therapies by simultaneously targeting both the cell cycle and PI3K signaling pathways. Clinical studies have shown promising efficacy in patients with heavily pretreated PIK3CA-mutant ER-positive HER2-negative advanced breast cancer as well as some activity in other solid tumors[1][7][5].
