PALI-0008

PALI-0008 is a highly potent, selective small molecule inhibitor of phosphodiesterase 4 (PDE4), developed as the active metabolite of the colon-targeted prodrug PALI-2108. It demonstrates strong inhibition of tumor necrosis factor-alpha (TNFα) production in human whole blood, with an IC50 of 0.022 μM, and exhibits approximately 20-fold higher potency than apremilast in reducing TNFα levels. PALI-0008 is designed to act locally in the colon following oral administration of its prodrug form, minimizing systemic exposure and central nervous system side effects commonly associated with other PDE4 inhibitors. The drug is being developed by Palisade Bio for inflammatory bowel diseases such as ulcerative colitis and fibrostenotic Crohn’s disease[1][4][7].
