Overview

PALI-1908 is a microbiota-activated prodrug of a phosphodiesterase 4 (PDE4) inhibitor, designed to be locally activated in the colon by bacterial enzymes. It leverages advancements from the related compound PALI-2108 and is being developed specifically for the treatment of fibrostenotic Crohn’s disease. The prodrug design aims to minimize systemic absorption until it reaches the colon, where it is bioactivated by colonic bacteria—potentially leading to targeted anti-inflammatory effects with reduced systemic side effects. The mechanism involves inhibition of PDE4, which increases intracellular cAMP levels and downregulates inflammatory cytokines and cell adhesion molecules, thereby reducing local inflammation in intestinal tissues[1][2][3].
