Overview

Palucorcel (CNTO 2476) is an investigational cell therapy consisting of human single-donor umbilical tissue-derived cells (hUTC). Developed by Janssen Research & Development (a Johnson & Johnson company), it was primarily evaluated for the treatment of retinal degenerative diseases, including geographic atrophy (GA) secondary to age-related macular degeneration (AMD) and retinitis pigmentosa. The therapy is administered via subretinal injection, often utilizing specialized delivery systems such as the iTrack microcatheter or a suprachoroidal surgical approach. Its mechanism of action involves neuroprotection and the rescue of retinal pigment epithelium (RPE) phagocytic function, which helps prevent the loss of photoreceptors. Clinical development reached Phase 2b for GA, but the program faced challenges due to high rates of surgical complications, such as retinal detachments and perforations, and a failure to show significant efficacy in reducing GA lesion growth.
