Overview

PAM-1 is a cathelicidin-derived antimicrobial peptide (AMP) originally identified in the platypus (*Ornithorhynchus anatinus*) genome. It is currently under preclinical investigation as a biologic therapeutic agent for multi-drug resistant bacterial infections, specifically targeting ceftazidime-avibactam (CZA)-resistant and carbapenem-resistant *Escherichia coli*. PAM-1 exerts its antibacterial effects by directly disrupting bacterial cell membranes and inhibiting biofilm formation. Additionally, it possesses immunomodulatory properties, demonstrating the ability to downregulate the expression of inflammatory cytokines. In vivo studies using the *Galleria mellonella* infection model have shown significant survival improvements (>50%), suggesting potential for further development against high-priority antibiotic-resistant pathogens.
