Overview

PAM-5129 is an orally bioavailable small molecule degrader targeting Interleukin-1 receptor-associated kinase 4 (IRAK4). Developed by Pamplona Biotech (潘普洛纳生物科技), it is part of the P001 program, which also includes a topical formulation (PAM-194). IRAK4 is a key mediator in the Toll-like receptor (TLR) and interleukin-1 receptor (IL-1R) signaling pathways, which are central to innate immunity and inflammatory responses. By degrading IRAK4 rather than simply inhibiting its kinase activity, PAM-5129 aims to more effectively block both the kinase-dependent and scaffolding functions of the protein. It is currently in the preclinical candidate (PCC) stage for the treatment of various autoimmune and inflammatory conditions, including systemic lupus erythematosus (SLE), inflammatory bowel disease (IBD), rheumatoid arthritis (RA), and atopic dermatitis (AD).
