Drug intelligence / Profile preview
PAM-II (also known as EPGN-1137) is a small-molecule positive allosteric modulator (PAM) of the alpha-7 nicotinic acetylcholine receptor (α7 nAChR). It is currently in preclinical development by Epigen Biosciences (in collaboration with the University of North Texas Health Science Center) for the treatment of stroke and traumatic brain injury (TBI). By positively modulating α7 nAChR, PAM-II enhances endogenous cholinergic pathways, which are widely expressed in neuronal and immune tissues, to promote neuroprotection and reduce neuroinflammation. This mechanism of action is also being explored for potential therapeutic extensions into cognitive and psychiatric disorders, such as dementias and schizophrenia.
