PAM Therapy is an enzyme replacement therapy (ERT) being developed by PAM Theragnostics for the treatment of Alzheimer's disease and the prevention of cognitive decline. The therapy utilizes the endogenous peptidylglycine alpha-amidating monooxygenase (PAM) enzyme to compensate for PAM deficiency, which has been observed in the cerebrospinal fluid of Alzheimer's patients. PAM is the sole enzyme responsible for the C-terminal amidation of over 70 bioactive peptide hormones, including vascular protective peptides like adrenomedullin. By restoring PAM activity, the treatment aims to normalize peptide hormone balance and restore blood-brain barrier (BBB) functionality. The drug is designed for weekly subcutaneous administration, with PEGylated versions showing sustained activity in circulation for up to seven days.
