Overview

Pam2CSK4 is a synthetic diacylated lipopeptide that acts as a potent and selective agonist of the Toll-like receptor 2 (TLR2) and Toll-like receptor 6 (TLR6) heterodimer. It mimics the acylated amino terminus of bacterial lipoproteins, triggering innate immune responses by activating NF-κB and AP-1 transcription factors, leading to cytokine secretion and inflammatory signaling. Pam2CSK4 has been used extensively in research as an immunostimulatory agent, adjuvant for vaccines (notably enhancing humoral and cellular immunity in SARS-CoV-2 nanoparticle vaccine studies), and for studying platelet activation via TLR-mediated pathways. Its mechanism involves cooperation with TLR6 to mediate immune cell activation, promote apoptosis in response to lipid moieties of lipoproteins, induce platelet aggregation/adhesion, and stimulate pro-inflammatory cytokine production[1][5][6][7].
